| I. Background | |
| PD-1（程序性死亡受体 1），T 细胞表面的一种免疫检查点受体，通常用于抑制 T 细 胞过度活跃，但癌细胞会利用它来逃避攻击，导致 T 细胞“耗竭”。而 IL-2 作为 T 细胞增 殖与存活的关键细胞因子，可扩增效应 T 细胞（Teff）及自然杀伤细胞（NK），但传统 IL -2 同时激活调节性 T 细胞（Treg），可能削弱免疫应答。 将 PD-1 检查点阻断与 IL-2 对 T 细胞的增强作用相结合，从而创造出能够“唤醒”肿 瘤内耗竭 T 细胞的疗法，以实现更强、更精准的抗癌反应，且潜在毒性可能低于单独使用 这两种物质。这些分子能够将 IL-2 直接递送至表达 PD-1 的肿瘤浸润淋巴细胞（TILs）， 增强其杀伤癌细胞的能力。 |
