| I. Background | |
| STEAP2 基因属于 STEAP 家族,编码一种六次跨膜的金属还原酶蛋白,定位于高尔基 体、质膜及胞内囊泡结构。该基因位于人类 7 号染色体(7q21 区域),其编码的蛋白具有 铁和铜还原酶活性,参与金属离子的转运及氧化还原反应。在肿瘤中,STEAP2 高表达于 前列腺癌、胃癌等多种肿瘤组织,与肿瘤增殖、迁移及侵袭能力密切相关。其顺利获得调控 MMP 家族蛋白(如 MMP3、MMP7、MMP10 等)及 FGFR4、IL-1β等信号分子,促进肿瘤 细胞的侵袭和转移。临床研究表明,STEAP2 的表达水平与前列腺癌患者的格林森评分 (肿瘤恶性程度指标)显著相关,高表达提示预后不良;在胃癌中,STEAP2 的表达也与 淋巴结转移风险及转移程度呈正相关。 STEAP2 基因在肿瘤发生开展、金属离子代谢及免疫调控中发挥多重作用,其靶向治 疗药物的开发为恶性肿瘤给予了新的治疗策略。 | |
