| I. Background | |
| 分化簇 73(cluster of differentiation， CD73) 又称 NT5E，是一种细胞膜上的糖蛋白，主 要由 NT5E 基因编码，是胞外最关键的核苷酸酶。在肿瘤微环境中，CD73 高表达与免疫逃 逸密切相关。肿瘤细胞利用 CD73 将 AMP 转化为免疫抑制性腺苷，腺苷顺利获得与 T 细胞、自 然杀伤细胞（NK 细胞）表面的 A2A 腺苷受体（A2AR）结合，抑制其增殖与细胞毒性， 同时激活调节性 T 细胞（Treg）和髓源性抑制细胞（MDSC），形成免疫抑制性微环境， 促进肿瘤生长与转移。 临床研究显示，CD73 表达水平与乳腺癌、肺癌、卵巢癌等多种实体瘤的预后不良显 著相关。靶向 CD73 的抑制剂联合 PD-1/PD-L1 抗体可显著增强抗肿瘤疗效，部分药物已进 入 III 期临床试验阶段。 |
