| I. Background | |
| Notch1 作为 Notch 受体家族核心成员，由胞外配体结合域（EGF 重复序列）、跨膜域 及胞内域（NICD）组成。当 DLL4 结合 Notch1 胞外域后，引发两步蛋白酶切割：ADAM 蛋白酶（如 ADAM10/17）切割胞外段，随后γ-分泌酶切割跨膜区，释放 NICD 进入细胞核。 NICD 与转录因子 CSL 及辅激活因子 MAML 形成复合体，激活下游靶基因（如 Hes1、 Hey1、Myc），调控干细胞维持、细胞增殖及分化。 DLL4 与 Notch1 是 Notch 信号通路 的关键组分，顺利获得细胞间接触依赖性信号调控发育、 血管生成及疾病进程。DLL4 是 Notch 配体家族成员，作为跨膜蛋白特异性地结合并激 活 Notch1 受体，驱动细胞命运决定、组织稳态及病理重塑。 |
